Trevigen’sGlutathioneAssayutilizesacarefullyoptimizedenzymaticrecyclingmethod,whichemploysGlutathioneReductase,forthequantificationofglutathione.ThesulfhydrylgroupofglutathionereactswithDTNB(5,5’-dithiobis-2-nitrobenzoicacid,Ellman’sreagent)andproducesayellow-colored5-thio-2-nitrobenzoicacid(TNB).Themixeddisulfide,GSTNB(betweenGSHandTNB)thatisconcomitantlyproduced,isreducedbyGlutathioneReductasetorecycletheGSHandproducemoreTNB.TherateofTNBproductionisdirectlyproportionaltotheconcentrationofGSHinthesample.Thus,measurementoftheabsorbanceofTNBat405or412nmprovidesanaccurateestimationoftheamountofglutathioneinthesample.
