Trevigen’skithasanoptimizedTUNEL-basedsystemthatpreferentiallylabelsthedouble-strandedDNAfoundinapoptoticcells.DNAfragmentsgeneratedduringapoptosisareend-labeledwithmodifiednucleotidesusingahighlypurifiedterminaldeoxynucleotidyltransferaseenzyme(TdT)inauniquenon-toxiclabelingbuffersupplementedwithatailoredcationtomaximizethe signal-to-noiseratio.TheincorporatednucleotidesaresubsequentlydetectedusingahorserADIshperoxidaseconjugate.Theconjugatecatalyzestheconversionofdiaminobenzidine(DAB)intoavisIBLedarkbrownprecipitate.ThekitincludesmethylgreencounterstainwhichisageneralcounterstainappropriateforawidevarietyoftissuesandtumortypesandprovidesgoodcontrastwiththebrownDABinlabeledcells.ADABenhancerisalsoprovidedwiththekitfortheoptiontointensifyordarkenDABstaining.
